I would think that EWR-HKG on CO has to be up there. After all, it was the world's longest non-stop flight until SQ started LAX/EWR to SIN.

JNB-ATL is 7379NM and an elapsed time of 17:45hrs with SAA A346 while HKG-EWR is 6994NM and 15:45hrs elapsed time for CO with B772. SIN-LAX is numero uno though.
