I was thinking about taking a spring trip to visit friends who live near LEJ. I have been there before by taking DL (NW before that) to FRA then having another ticket on LH. That is a pain in the butt because I have to collect my luggage in FRA and check in again to go to LEJ. For a time I could fly AF DTW-CDG-LEJ, which was great because I could stick with Skyteam the entire trip.
The easiest, along with the cheapest, way to get there is LH DTW-FRA-LEJ, but since I am going that far I would like to earn as many Skyteam miles as possible, since I usually fly DL.

PRG also isn't very far from LEJ, about 150 miles (250 km) by road, 3 hr. drive or 4 hrs. by train (looks like you have to change trains at DRS). Not sure if there any other ground transportation options such as bus. DL operates nonstop daily JFK-PRG in the summer, starting May 22 I believe. Or KL via AMS or AF via CDG if you want to use SkyTeam carriers.

If I may - no, that's the problem: You would have to take the Airport Express Bus from TXL to Berlin main station.
Other than that, DAL763ERs suggestion isn't bad at all.
The so called ICE-train takes you to Leipzig in 1h12m...

It's a little pricey, but if you are looking for variety and unique entries for your flight log, you might consider flying DTW-AMS and then connecting to Darwin/Etihad Regional that flies AMS-LEJ on the Saab 2000.
If it were me, I'd seriously consider flying into PRG and out of TXL, taking trains in between. Or something similar. DL usually prices open-jaw tickets like that about the same as the round-trip price to either city, and I like to take advantage of that opportunity to see some different scenery. And I've been waiting half my life for a chance to visit PRG...
