What Happens Between First And Second Flight?

With a number of all new airliner designs taking to the skies - one hopes - for the first time in 2008 and 2009 (eg Superjet, 787, ARJ21), I was wondering what happens after the first flight. What is checked before the second flight takes place? What bearing does information gained from the first flight have on what is undertaken in the second flight?
