They could, but by the time the timings would be sub-optimal. Many Porto travellers will want an early morning flight to LIS. Operating the pattern you suggest and it would probably be a mid-morning flight. Using a Madrid based aircraft would be a bit better, but if Easyjet wanted the day trip market then they would probably need a Portugese based aircraft
However could a 1 aircraft operation be done using nightstopping crews, as happenned at FCO initially and will happen at DSA this summer?

LIS-OPO is not a profitable route for TP, according to the interview published today in "Expresso". The route serves primarily as a connecting link to TP's LIS hub. In any LCC wants to get in on it I don't think TP would have any problem with it. Personally I doubt that even a LCC could make any money on it. The 2 cities are just 300Km apart and linked by 2 highways, express bus service and express trains.
