There are lots of programs out there.

Burt Rutan's SpaceShipOne has seen the greatest success but there are others out there.

Bigelow's Simplified Orion

Which ones could really have revolutionary effects on the industry and long term influence on how we explore the heavens?

Yes they have a lot of potential especially with their NASA contract.

It will be used, right?

I wonder if we could ever see runway-space-runway spacecraft in the next decade or so. Rutan is close and so is SpaceDev. Xcor is also working on this concept.

I feel it would be a cheaper and safer option compared to any rocket with capsule option.

I believe that once it gets going EADS Astrium has good chances of becoming important.

If I was to buy a Space flight for myself, I would buy it from them rather than from Virgin Galactic.
They team with Arianespace so their Space transport technology must be top-notch.  airplane 
There was a better way to fly it was called Concorde

But no cigar. Flying at 1000 m/s is not the same thing as flying at 7500 m/s. Rutan is nowhere close to achieving orbit-- in fact, he's not even trying.

Which will be the first private company to to space, go into orbit and land on a runway and turn around in 2 weeks.

It seems the X-prize was great, but never upped on itself for the next prize...orbital flight.
Would love to see someone create a spacecraft that launches from a runway and lands on a runway and can then do it again in 2 weeks.

Who is closest on this?

I would think that eventually private industry will go this way for paying travelers. Do you think SRB will be satisfied with just sub-orbital?

Rutan could probably get to orbit with a variation of his SS1, maybe as a third stage of a much larger system, but getting back down is another story. In the real world, noone is anywhere near as far alond as Elon and his Dragon. NASA is insisting on new Dragons for each cargo flight, so SpaceX should have plenty of pre-owned capsules to use in a few years.
