My guess would be American or Northwest. Northwest has got a good route network to central and western Canada via their Minneapolis hub. American has got a good amount of flights from New York, Boston, Chicago, and Dallas to major cities throughout Canada.

AA I know flies to YHZ, YQB, YUL, YOW, YYZ, YYC, and YVR, and I'm not sure, but they may fly to YWG and YEG as well. Northwest, I'm not so sure about, but I know they fly to YYZ, YWG, and YYC. I'm sure there are more flights.

Don't forget Horizon Air, which serves YYJ, YVR, YYC, YEG, and YLW.

Well, since United codeshares with Air Canada, the answer for the US airline with the most flights *should* be United.

Northwest has the most destinations served by its own aircraft (including NWExpress carriers):

YUL, YOW, YYZ, YXU, YQT, YQK (seasonal), YWG, YXE, YQR, YYC, YEG, YVR and possibly a couple more.

Any Canadian cities not served by any of the U.S. majors that have potential to be served?

I do not think that AA serves YWG - they have started and stopped it at least twice.

AA no longer flies to YHZ either. I'm pretty sure for own-aircraft flights and capacity (i.e. mainline & regional partners, not codeshare), NW is 1st, AA 2nd and DL 3rd.
