It depends on what airline you shall be flying.
The authority for jumpseat rides resides with the Captain, so he or she is the person you should place your request to.
A member of the flightcrew is usually at the door for boarding, so this your best bet.
I got the jumpseat once on Air India, Boeing 747-337 "Shivaji" (don't remember the reg lol..). I had my request turned down by the purser, but I got lucky, the Captain was present and granted my request.
Now it also helps if you're a pilot lol
