Do airlines station backup aircraft at their major hubs in case an airplane breaks down and they suddenly need a replacement?

NW has a backup DC10 in AMS (only for the summer).

Yes. Most try to. A lot of the spares are used for charters, especially
during the NFL season. There are times that due to maintainence,
damage, airworthiness directives, etc, the spares are not available.

Does EasyJet or Ryanair have any back up aircraft, if so how many?

I think FR and U2 rely a bit on Titan Airways and Monarch for back up, but I could be wrong.
