B757-200 Freighter v/s A321P2F

Can the recently converted STC on A321P2F be the near best replacement for the older B757-200 Freighters.

Yes, the A321 freighter is the closest you will find to a 757-200F. Slightly more volume, slightly less payload and less range (but most 757Fs don't fly very long routes anyway)

The STC is the "Supplemental Type Certificate". You don't convert the STC. The STC is the piece of paper that allows you to convert the A321.

I believe thrust on the powerplant would be less for the A321P2F

The 757's runway/engine performance has been praised quite a lot on here (and other sites). Are there 757 routes where A321P2F/737F would struggle on?

Not that I can think of. All the 757 freighters I see do relatively short flights between airports with adequate runways for an A321. I looked up a random smattering of FedEx 757s on FR24. The longest flight I could find was a bit over 4 hours, but the vast majority are only around 1 hour. Same story at DHL. Blue Dart in India is even less, a max around 2 hours and most don't even make 1 hour.
The A321 isn't known for being runway restricted.
